Initiator caspases

WebbInitiator caspases (e.g., CASP2, CASP8, CASP9, and CASP10) cleave inactive pro-forms of effector caspases, thereby activating them. Effector caspases (e.g., CASP3, CASP6, CASP7) in turn cleave other protein substrates within the cell, to trigger the apoptotic process. The initiation of this cascade reaction is regulated by caspase inhibitors. WebbIndividual caspases differ in their substrate specificities and are subdivided into three distinct groups. The sequence preferences generally correlate with caspase function as apoptotic initiators (caspases-2, 8, 9, and 10), executioners (caspases-3, 6, 7), and cytokine processors (caspase-1, caspase-11; 15–19).

Webb2 sep. 2008 · The initiator caspases appear to be highly specific proteases that cleave few proteins other than their own precursors and the downstream effector caspases (3, 4). Thus, the bulk of the proteolysis that takes place during apoptosis is carried out by the effector caspases. Webb1 mars 2015 · Initiator caspases (caspases-2, -8, -9 and -10) exist as latent zymogens that gain proteolytic activity upon their recruitment into multi-protein platforms that promote their proximity-induced autoactivation such as the death-inducing signalling complex (DISC) that activates caspases-8 and -10, and the apoptosome that drives caspase-9 …
